Mitchell scored his third try in the 58th minute when he bolted away 60m untouched after juggling a pass from Mumm. Barnes converted to make it 45-5.
Four minutes later, Mitchell continued to entertain, when he finished off another long-range effort.
A consolation try to replacement Lions forward JC Janse van Rensburg did little to dim the pain for the ordinary visitors.
Two tries in the space of three minutes from Holmes nudged the Waratahs beyond 60-points, making it 66-12 by the 77th minute.
Incredibly, the scoring was only completed after the full-time siren when Turner picked up a stray kick, racing 90m to score under the posts.
The Waratahs are away to the Western Force in Perth next Saturday night, with the Lions still searching for their first win of the year when they play the Crusaders in Christchurch.
