The Mercedes man regained the lead in the drivers' standings by winning in Malaysia last time out and he is aiming to cement top spot at the Japanese Grand Prix on Sunday.
The Briton won on his first outing in Japan when racing for his old team McLaren in 2007, but that victory came in a rain-affected race at the Fuji Speedway.
His best effort at the famous Suzuka circuit was a third-placed finish in 2009 - the same year the track was reinstated to the F1 calendar.
The 29-year-old now wants to add to a list of winners that includes F1 greats such as Michael Schumacher, Ayrton Senna, Mika Hakkinen and Sebastian Vettel.
"I've never won at this circuit and have only made the podium once, on my first visit way back in 2009," Hamilton said.
"This year, though, we have an exceptional car and I'm really hoping I'll finally have my shot at the top step.
"All the greats of Formula 1 have won at Suzuka since the sport first came there in the 1980s and I'm determined to add my name to that list this weekend."
Hamilton, who won the title in 2008, leads the standings by three points from team-mate Nico Rosberg.
